Synthesis of phosphono-bis(phosphinate) cyclam derivative

Radiopharmaceuticals containing copper isotopes can be used as diagnostic tools (methods PET, SPECT) or agents for tumour treatment. Copper radionuclides must be applied into the organism in the form of a coordination compound with suitable ligand. Derivatives of 1,4,8,11-tetraazacyclotetradecane (cyclam) are some of the most suitable ligands for Cu (II). A coordination compound must be thermodynamically stable and kinetically inert, the ligand must coordinate the metal ion as quickly as possible. In this Diploma thesis, one of the first unsymmetrically substituted derivatives of cyclam has been synthesized using two methods of orthogonal protection. The synthesized ligand contains one phosphonate and one methylene bis(phosphinate) pendant arm. Copper complex of the ligand was prepared, and its acid assisted dissociation kinetics were studied.
UV-Vis and IR Spectroelectrocbemistry of Copper Complexes and Bioactive Compounds

The interpretation of the change of absorption spectrum of an oxidized and reduced molecule\nrecorded during the electron transfer is an efficient tool for the determination of oxidation or\nreduction mechanism. This technique provides the information about the electroactive\nchromophore and is performed in two regimes of electrochemical measurement, cyclic\nvoltammetry and chronoamperometry, respectively. This approach was successfully applied to\nstudy the fundamental electrochemical behavior of recently synthesized copper complexes\nproviding biomimetic activity, ofpolyphenolic bioactive compounds, and also for the reduction\nof halogenated phenanthrolines. The identification of redox products was done by\nchromatographic techniques as HPLC-DAD and HPLC-ESI-MS/MS.
Ligand design for medicinal applications

In recent years, copper radioisotopes have been extensively studied for their suitable coordination and physical properties. Nuclides 61 Cu, 64 Cu and 67 Cu are used in nuclear medicine - in diagnostic as well as in therapeutic applications. The aim of the Thesis is a study of the coordination properties of divalent copper as a stepping stone for the next potential applications. The presented Thesis consists of two thematic parts. The first part deals with the synthesis of cyclam derivatives. Sixteen new macrocyclic ligands with different phosphorus acid coordinating pendant arms (phosphinate, phosphonate, germinal P-C-P) were prepared; an analogous ligand endowed by carboxylic acid pendant arm as well as tetramethylcyclam without coordinating arm were prepared for comparison. The influence of the nature of coordinating acid pendant arms on selectivity and on the rate of copper(II) complexation was studied in detail. The protonation constants of the free ligands and the stability constants of the complexes with selected transition metal ions were determined by potentiometric titrations and by 1 H and 31 P NMR spectroscopy. Synthesis of asymetrically substituted derivatives of cyclam

Copper radionuclides have a potential in nuclear medicine as diagnostic or therapeutic agents. Copper isotopes are used in the form of coordination compounds with various ligands, such as derivatives of 1,4,8,11-tetraazacyclotetradecane (cyclam). Properties of such complexes can be modified by pendant arms introduced on amino groups of cyclam. This thesis is aimed on the synthesis of asymmetrically substituted cyclam derivative with derivatives of phosphorous acids in 1,8- position. In spite of the fact, that the target ligand has not been successfully synthesised, various possibilities of substitution on diprotected cyclam were investigated. Use of Monovalent Copper for Sensitive Detection of Methyl Derivatives of Xanthine

This work is concerned with the electroanalysis of xanthine (Xan) and its methylated derivatives (1-, 3-, 7-, and 9-mXan) on a pencil graphite electrode in the presence of copper ions. The main idea of using copper consists in the in situ formation of the complex Cu(I)-mXan on the electrode surface and improvement of oxidative signals of the corresponding mXan during the detection. Linear sweep voltammetry (LSV) in connection with adsorption stripping techniques was used for the determination of mXan. To enhance the oxidation signals the elimination voltammetric procedure (EVP) was applied.

Spectroelectrochemistry of Flavonolignans and their DNA-binding Copper Complexes

The oxidation of flavonolagnan silybin and its selected derivatives was studied on glassy carbon electrode in aqueous environment by means of cyclic voltammetry, in situ UV/VIS spectroelectrochemistry and absorption spectrometry. The oxidation mechanism of flavonolignans and their complexion with copper was investigated under anaerobic conditions. The acquired results were interpreted in context of protective (antioxidant) and DNA-binding (prooxidant) properties of flavonolignans and their transition metal complexes.
